Bt2-Ins(1,4,5,6)P4/PM is a membrane-permeant, bioactivatable derivative of myo-inositol 1,4,5,6-tetrakisphosphate (Ins(1,4,5,6)P4).
The negatively charged phosphate groups are masked by propionoxymethyl (PM) ester groups, while additional butyryl ester groups increase lipophilicity and facilitate cellular uptake. Following entry into cells, intracellular esterases cleave the ester groups, enabling intracellular delivery of the corresponding inositol tetrakisphosphate.
Bt2-Ins(1,4,5,6)P4/PM has been used as a membrane-permeant inositol tetrakisphosphate analog in studies of calcium signaling, chloride transport and inositol phosphate-dependent cellular processes. It is also suitable as a positional-isomer control in comparative studies with other membrane-permeant InsP4 derivatives.
