INO-4995 is a membrane-permeant synthetic analogue of D-myo-inositol 3,4,5,6-tetrakisphosphate (Ins(3,4,5,6)P4). The compound is esterified to facilitate cellular uptake and is converted intracellularly to the less membrane-permeant active metabolite INO-4913.
INO-4995 has been investigated as a modulator of epithelial ion transport. Studies in cystic fibrosis airway epithelia demonstrated effects on epithelial Na+ transport and fluid absorption, while subsequent work investigated its effects on Ca2+-activated chloride conductance and TMEM16A/ANO1.
INO-4995 is suitable for research on inositol phosphate signaling, epithelial ion transport, ENaC regulation, Ca2+-activated chloride channels and cystic fibrosis-related cellular mechanisms.
