Membrane-permeant myo-Inositol-1,4,5-Trisphosphate Derivative
Bt3-Ins(1,4,5)P3/AM is a membrane-permeant, bioactivatable derivative of myo-inositol 1,4,5-trisphosphate (Ins(1,4,5)P3, IP3), designed to enable intracellular delivery of this important second messenger. It is a useful tool for studying IP3 receptor-mediated signaling and intracellular calcium mobilization in intact cells.
The negatively charged phosphate groups are masked by acetoxymethyl (AM) ester groups, while additional butyryl ester groups increase lipophilicity and facilitate cellular uptake. Following entry into cells, intracellular esterases cleave the ester groups, enabling intracellular delivery of biologically active Ins(1,4,5)P3.
Bt3-Ins(1,4,5)P3/AM can therefore be used to investigate IP3 receptor activation, intracellular Ca²⁺ signaling and downstream cellular responses without direct microinjection of Ins(1,4,5)P3. Applications include cellular signaling studies, calcium flux experiments and experiments combining intracellular IP3 delivery with live-cell Ca²⁺ imaging.
